Mechanism study on the improvement of inflammation in rat glomerular mesangial cells by regulating the JAK2/STAT3 pathway by Yi shen Yang Yin Compound-containing serum

Systemic lupus erythematosus (SLE) is a common autoimmune disease that usually involves multiple organs and systems throughout the body. Among them, lupus nephritis (LN) has a prevalence of about 50-70% in SLE patients. Currently, glucocorticoids, immunosuppressants, biological agents and other treatments are mainly used for LN patients, but long-term application of patients can develop tolerance, However, long-term use of these drugs can lead to tolerance, adverse reactions, and decreased efficacy. Therefore, new therapeutic strategies are needed to treat or delay the progression of LN. In the preliminary clinical trials, we found that Yi Ren Nourishing Yin Combination has better efficacy on LN patients in terms of hormone withdrawal and reduction of urinary proteins; in order to further confirm the effect, we found that Yi Ren Nourishing Yin Combination can improve renal function and renal pathology through animal experiments in MRL/lpr mice (mice with spontaneous lupus erythematosus nephritis); in order to explore the in-depth mechanism of the formula, we found that Yi Ren Nourishing Yin Combination can suppress JAK2 / STAT3 signaling through protein microarray screening. JAK2 / STAT3 signalling pathway activation, reduce Th17, IL-17 expression. On the basis of this project, we will use the serum containing Yi Ren Nourishing Yin Compound to intervene in the inflammation model of rat glomerular membrane cells, and detect the cell proliferation, cell cycle and expression of related molecules by blocking the JAK2/STAT3 pathway, so as to explore the regulation and mechanism of the formula on the JAK2/STAT3 signalling pathway and its downstream molecules at the cellular level.
