Effects and molecular mechanisms of PDCD4 regulation by miR-195a-5p in rats with osteoarthritis of the knee by Shenwei Gubi Tablets

Abstract

Osteoarthritis of the knee (KOA) is a degenerative joint disease with a high incidence in the middle-aged and elderly population. The main clinical features include knee pain, impaired movement and muscle dysfunction, which seriously affects patients' quality of life. Shenwei Gubi Tablets are mainly composed of Wei Ling Xian, Xiao Hong Ginseng, Bai Shao, Huai Niu Kne and Dou Wu, which are effective Chinese medicinal preparations for the clinical treatment of osteoarthritis in Kunming Chinese Medicine Hospital. Previous studies have confirmed that Shenwei Gubi Tablets can effectively reduce the TNF-α content in the joint fluid of rabbit knee osteoarthritis model and inhibit the inflammatory response. The expression of miR-195a-5p in chondrocytes was significantly increased after stimulation with Shenwei Gubi Tablets, and a dose-dependent trend was observed. The expression of mRNA and protein of inflammatory factors and PDCD4 in chondrocytes was significantly decreased after stimulation with Shenwei Gubi Tablets, and the apoptosis rate was significantly decreased, while the cell survival rate was significantly increased, and a dose-dependent trend was observed. Shenwei Gubi Tablets inhibited the inflammatory response of the KOA cell model through miR-195a-5p regulation of PDCD4, thereby alleviating the degeneration of cartilage under the pathological effect of OA. This study reveals the specific mechanism of Shenwei Gubi Tablets in the treatment of KOA and provides a more solid theoretical basis for the application of Shenwei Gubi Tablets in clinical treatment.
